Human hepatic clearance from in-vitro CLint, with the error band measured on known drugs.

Overview

AI-generated overview

Remote MCP server for ADME/IVIVE calculations in early drug discovery: predicts human clearance inputs and hepatic clearance from SMILES and in-vitro data.

What you can do
  • Predict fraction unbound and unbound intrinsic clearance from a SMILES, with structure-only CL and Vss estimates
  • Calculate hepatic and plasma clearance from in-vitro CLint, fu,mic and fu,p, with an observed-CL band and applicability warnings
  • Back-calculate the in-vitro CLint an assay must read to hit a target plasma clearance
  • Show how far plasma clearance moves as each input is varied over a range
  • Find nearest reference drugs by structure, with measured human plasma CL and the IVIVE prediction from their measured inputs
  • Predict human plasma CL from observed animal in-vivo CL (rat, dog, monkey) using every applicable method and its measured error
When to use it
Useful for medicinal chemists and DMPK scientists who want an assistant to run quick clearance and IVIVE estimates, set CLint cut-offs for screening cascades, or sanity-check how much to trust a prediction for a given chemotype.
Requirements
A remote MCP endpoint reached over streamable HTTP; no packages, environment variables or headers are declared. Network access to the provider's API is required.
Before you install
Results are research estimates and the server says they are not for clinical, regulatory or dosing decisions. Predictions from predict_inputs are not measurements, and downstream results built on them rest on predicted inputs; the server asks that warnings and caveats be relayed. Compound structures (SMILES) are sent to the provider's API.

Installation

In SourceWeft

  1. Open ivive.app in the dashboard and add it to a workspace.
  2. Enable the server for the chats that should use its tools.

Web executable via Streamable HTTP. Remote servers run from the web runtime once configured in a workspace.

Use with an agent

Paste this prompt into Claude Code, Codex, Cursor or another agent — or into SourceWeft chat. The agent reads this server's install guide, shows you its publisher, endpoint and sign-in, and adds it to your client once you agree. Keys never go into a command.

Read https://sourceweft.com/mcp/app.ivive%2Fivive/install.md and connect the MCP server it describes to my client. Before changing any config, show me its publisher, endpoint and sign-in method and wait for my OK. Never put an API key in a command or in a file I would commit; ask me where it should come from.

Read the install guide the agent follows

Tools

6

predict_inputs

Predict fu,p and fu,inc from a SMILES, with structure-only human CL and Vss as second opinions. Predictions, not measurements; check the applicability verdict.

Parameterssmilesrequiredassay_typeconcentration

calc_clhep

Hepatic and plasma clearance from in-vitro CLint, fu,mic and fu,p (IVIVE), with the observed-CL band and applicability warnings. If fu,p or fu,mic came from predict_inputs, tell the user the result rests on predicted inputs.

target_clint

The in-vitro CLint an assay must read for a compound to reach a target plasma clearance (mL/min/kg): calc_clhep run backwards, e.g. to set a CLint cut-off for a screening cascade. Needs fu,mic and fu,p; refuses a target at or above the liver blood flow ceiling and returns that ceiling. Uncorrected IVIVE, so no error band.

input_sensitivity

How far plasma CL moves as each input is varied over a range, e.g. a predicted fu,p over its 50% range.

similar_drugs

Nearest reference drugs to a SMILES (Tanimoto, Morgan radius 2), each with measured human plasma CL and the CL IVIVE predicted from its measured inputs. Use it to judge how far to trust calc_clhep for this chemotype: if IVIVE missed on the neighbours, expect it to miss here. Returns neighbours only, not a prediction for the query.

Parametersnsmilesrequired

scale_to_human

Predict human plasma CL from observed animal in-vivo CL (rat, dog, monkey), every applicable method with its measured error. Observed animal CL only, never predicted.
